He lies to Othello about where Desdemona's handkerchief came from. Iago tells him that Cassio had it where in reality Emilia had taken it from her. This poisoned Othello and sent him into a fit of rage and jealousy. Iago also gets Cassio talk to Desdemona about him getting his position returned to him. This made it look like the two of them were spending intimate time together to further push that Desdemona was cheating on Othello with Cassio.
One of Iagos lies is that desdemona is cheating on othello. This poisons othello's marriage by filling him with jealousy which will eventually push him to a breaking point.
